Enzyme-microwave assisted extraction of polysaccharides from the adventitious root of Atractylodes chinensis and evaluation of hypoglycemic potential

Atractylodes chinensis (DC.) Koidz is a medicinal herb known for its significant therapeutic potential, and polysaccharides are one of its main bioactive ingredients. Despite their importance, optimal extraction methods for these polysaccharides require further investigation. This study employed enzyme-microwave-assisted extraction (EMAE) to extract A. chinensis adventitious roots polysaccharides (AARP), and their hypoglycemic activity was evaluated. The extraction process was optimized using response surface methodology with Box-Behnken Design (RSM-BBD). Under the optimized conditions (cellulase concentration of 208 U/mL, enzyme incubation temperature of 57 °C, liquid-to-solid ratio of 46 mL/g, and microwave time of 6 min), the extraction yield of polysaccharides reached 23.08%. The purified AARP, obtained through macroporous absorption resin and gel-filtration chromatography, contains 70.64% carbohydrates and a notably high uronic acid content (12.47%). The molecular weight of AARP was determined to be 6.0 × 103Da, and it consisted of nine distinct monosaccharides, such as glucose, arabinose, and galactose, etc. AARP exhibited inhibitory effects on α-amylase and α-glucosidase, with IC50 values of 0.84 mg/mL and 0.68 mg/mL, respectively, indicating its potential as a hypoglycemic agent. This study not only established an efficient EMAE method for extracting hypoglycemic AARP but also enhanced the understanding of polysaccharide extraction from medicinal plant roots.
