IoT-based fully automatic smart providing aseptic materials for vitro and cryogenic collections of wild Juglans regia L. for genetic resources preservation

Walnut (Juglans regia L.) is one of the most economically important nut crops. In Kazakhstan, the natural J. regia population is registered in the south of the country and is characterised by a high polymorphism. Excised embryonic axes were used as explants for in vitro culture initiation and cryopreservation. Driver and Kuniyuki medium containing 4.4 μM of 6-benzylaminopurine, 0.04 μM indole-3-butyric acid was used to initiate in vitro culture. Of the 196 axes cultured, only 3% showed visual contamination. 80% of axes were viable and aseptic and were then micropropagated. These accessions from Kazakhstans J. regia population have never been cultured before, so this study contributes to the preservation of important Juglans genetic resources. Desiccation of the excised axes to 7.2% moisture content and further direct immersion in liquid nitrogen resulted in 76.7% recovery, which makes it possible to recommend this method for cryopreservation of the wild J. regia germplasm.
