Experimental Study of Nail Plate Implants for Keratoplasty

Introduction. One of the main disadvantages of artificial materials is that they can encapsulate, reject and disrupt the trophism of the cornea. In this regard, it seems justified to use a fragment of the nail plate, which is close in chemical composition and physical properties to the corneal tissue. Purpose of the study. To evaluate the biocompatibility of nail plate implants with corneal stromal tissue. Materials and methods. For analytical work, 5 samples of fragments of the nail plate were used; 4 of them were mechanically processed, the 5th – with the help of laser grinding. The following research methods were used: the method of culturing samples with a keratocyte cell line, the method of studying early apoptosis, the method of studying DNA comets, the method of scanning electron microscopy. Results. A significant growth of cell cultures was revealed in the presence of the studied samples of fragments of nail plates treated by mechanical and laser methods. Samples of fragments of nail plates treated by mechanical and laser methods, as implantation objects, do not trigger mechanisms of apoptosis in corneal keratocytes along the caspase and mitochondrial pathways. The mutagenicity of fragments treated by both methods does not exceed acceptable physiological values. The surfaces of the samples of fragments of the nail plate treated by laser method are ultrastructurally different from those treated by mechanical means: they have a smoother surface, no furrows from mechanical action. Conclusions. The conducted studies revealed the absence of toxic effects of nail plate implants and they are biocompatible with corneal stromal tissue.
