Quantitative analysis of hemopoetic chimerism after living-related homogeneous hemopoetic stem cells transplantation

BACKGROUND: Chimerism is a condition where genetically different cells coexist in the same organism. There are several states of chimerism, if after transplantation all blood cells are replaced by donor cells, it is possible to determine the full donor chimerism. It takes time, sometimes quite a long time, to achieve complete donor chimerism. The determination and study of chimerism allowed us to evaluate graft engraftment, as well as the patients condition in the post-transplant period, which was the main goal of our study. METHODS: Modern research methods are the basis for the implementation and optimization of methods for monitoring hematopoietic chimerism in the post-transplantation period of HSC. RESULTS: According to the results of our study on nosological forms, patients with acute myeloid leukemia prevailed in various variants 18 (58.6%). In this group, depending on the degree of HLAcompliance, a large proportion of patients had related ALLO-HSCTfrom donors selected 5/10, which was 8 pairs (44%). According to the results of the study, the information content of the amelogenin locus, which determines the number of pairs that do not coincide by gender, showed 62.5%. CONCLUSIONS: The information content of this locus was observed in 11 of the 14 heterogeneous pairs. In 3 pairs, when the donor-male/recipient-female combination was not observed, and full donor chimerism (99% of the presence of donor DNA) was noted according to the initial data of the definition of hematopoietic chimerism.
